摘要(Abstract):
16S rDNA基因克隆文库的建立和DGGE(变性梯度凝胶电泳)是分析油藏微生物群落结构和种群动态变化的重要方法,油藏微生物基因组DNA的提取是进行16S rDNA基因克隆文库的建立和DGGE的前提.建立大庆油田油藏微生物基因组DNA提取的方法,与基于SDS细胞裂解法相比,华舜基因组抽提试剂盒法具有DNA浓度高、纯度好和省时等优点.不同油藏条件下的实验和PCR扩增技术分析结果表明:该方法能够成功提取基因组DNA,并且提取的基因组DNA可以直接用于克隆文库和DGGE实验的细菌16S rDNA PCR扩增.这说明DGGE与分子克隆相结合的分子生物学方法在研究微生物提高原油采收率(MEOR)机理及指导MEOR在油田生产中的应用有着重要意义.
